The short answer is no. Manor Lords will be a single-player game and will not feature any multiplayer or co-op mode at launch, as per the official announcement by the dev Slavic Magic.

While answering the question about adding multiplayer to Manor Lords, Slavic Magic replied to a Redditor that they "want to polish the single-player element first before considering multi."

This means that we can't rule out the possibility of multiplayer or co-op mode being added to Manor Lords in the future; however, for now, you will have to play the game alone.
